Eine Lösung von Seidenfibroin (Bombyx mori) in 6 M Harnstoff wurde hergestellt, indem Seidenfibroin in 50% Lithiumthiocyanat-Lösung gelöst und durch Dialyse in eine 6 M Harnstofflösung überführt wurde. Durch diese schonenden Bedingungen wird die Spaltung von Peptidbindungen während der Auflösung vermieden.

Bei der Disk-Elektrophorese der Fibroinlösung in 6 M Harnstoff an Acrylamid-Gel bei pH 9,3 ergab sich eine Auftrennung in drei starke und einige schwächere Banden.

Durch Kurzzeit-Extraktion von Fibroin-Fasern mit Lithiumthiocyanatlösungen verschiedener Konzentration wurden lösliche Fibroin-Fraktionen erhalten, die eine vom Fibroin abweichende Zusammensetzung hatten.

Bei der Säulenchromatographie der Fibroinlösung in 6 M Harnstoff an DEAE-Cellulose wurde Fibroin in zwei Fraktionen aufgetrennt, von denen die kleinere Fraktion in ihrer Zusammensetzung stark vom Fibroin abweicht.

A solution of silk fibroin (Bombyx mori) in 6 M urea was prepared using conditions under which peptide bonds are stable.

By disc electrophoresis the fibroin solution was separated into three strong and several weak zones.

Short-time extraction of fibroin fibres with a solution of lithiumthiocyanate yielded a soluble fraction (4% by weight) with an amino acid composition, different from fibroin.

On DEAE-cellulose columns the fibroin solution in 6 M urea was separated into two fractions. The amino acid composition of one of these fractions differed markedly from that of fibroin.
