Summary
Cortisone, cortisole, and their synthetic derivates, prednisone, prednisolone, triamcinolone, and dexamethasone exhibit the following metabolic actions:
-
1.
Negligible inhibition of the DNA synthesis, on the other hand a strong inhibition of the RNA synthesis by all of the above corticoides.

2.
Blockage of the transport of nucleosides, and independent thereof, the transport of glucose into the cell by all of the various corticoids. Inhibition of the phosphorylation of nucleosides and nucleotides by cortisole, prednisolone and prednisolone-derivatives.

3.
In a cell free system, an inhibition of the messenger RNA synthesis is observed, due to the interaction of corticoides, given in a high dosis, with primer DNA.
All of these corticoids results, especially that of the fluoridated prednisolone derivatives, can be more clearly demonstrated on lymphatic leukemic cells, than on the Ascites cells.
In respect to their degree of cytostatic effectiveness on lymphatic leukemic cells, the tested corticoides can be arranged in the following order: The cytostatic effectiveness corresponds to the antiinflammatory and antiallergetic action on these substances which is in agreement with clinical experience.
These results can not be correlated to the action of corticoides as adrenal cortex hormones because they are observed under concentrations greater than the physiological norm.
Zusammenfassung
Cortison, Cortisol und deren synthetische Derivate Prednison, Prednisolon sowie Triamcinolon und Dexamethason zeigen an Ascitescarcinomzellen der Maus und an lymphatischen Leukämiezellen des Rindes in vitro folgende Stoffwechselwirkungen:

1.
Geringgradige Hemmung der DNA-Synthese, dagegen starke Hemmung der RNA-Synthese durch sämtliche geprüfte Corticoide.

2.
Blockierung des Nucleosid- und unabhängig davon des Glucoseeintritts in die Zelle durch sämtliche Corticoide. Hemmung der Phosphorylierung von Nucleosiden und Nucleotiden durch Cortisol, Prednisolon und Prednisolonderivate.

3.
Im zellfreien System wird bei hohen Corticoiddosen eine Hemmung der messenger-RNA-Synthese durch Wechselwirkung der Corticoide mit der PrimerDNA beobachtet.
Alle Effekte der Corticoide, besonders der fluorierten Prednisolonabkömmlinge, treten an lymphatischen Leukämiezellen wesentlich deutlicher hervor als an Asciteszellen.
Hinsichtlich der cytostatischen Wirksamkeit der getesteten Corticoide an lymphatischen Leukämiezellen ergibt sich folgende Reihenfolge: Cortison=Cortisol < Prednison=Prednisolon=6α-Methylprednisolon < Triamcinolon < Dexamethason. Die cytostatische Wirksamkeit entspricht damit der entzündungshemmenden, antiallergischen Wirksamkeit dieser Substanzen in Übereinstimmung mit den klinischen Erfahrungen. Die Effekte haben primär nichts mit der Wirksamkeit der Corticoide als Nebennierenrindenhormone zu tun, sondern werden bei wesentlich höherer Konzentration als der physiologischen beobachtet.
