Summary
Portal hypertension in rats was produced by placing Ameroid constrictors around the portal vein and the measurements of the portal pressure (intrasplenic pulp pressure) and liverportal venography were performed. 6 days after applying the constrictor the portal vein was occluded. The liver was only perfused by the hepatic artery and the blood from the splanchnic area was mostly shunted to the inferior caval system by preexisting dilatated anastomoses, only a part through oesophageal veins into the upper cava. The intrasplenic pulp pressure increased to a level twice of the normal range. In spite of the constant conditions in addition to the form of the portal vein block, this modell is useful to study the dynamic of the development and the pathophysiology of portal hypertension.
Zusammenfassung
Bei Ratten wurde durch Anlegen von Ameroidringen um den Pfortaderstamm ein protrahierter prähepatischer Pfortaderblock erzeugt und Druckmessungen (Milzpulpadruck) und Splenoportogramme durchgeführt. Bei diesem Modell kommt es regelmäßig zu einem völligen Verschluß der Pfortader nach 6 Tagen post operationem mit Ausschaltung der Leber aus dem Pfortaderstromgebiet und ausschließlicher arterieller Blutversorgung der Leber. Das Blut aus dem Splanchnicusbereich wird dabei im wesentlichen Über erweiterte präformierte portocavale Anastomosen zum unteren Hohlvenensystem abgeleitet, zu einem kleineren Teil Über Magen- und Oesophagusvenen zur V. cava superior. Gleichzeitig steigt der Milzpulpadruck auf mehr als das Doppelte der Norm an. Dieses Tiermodell ist auf Grund der konstanten Bedingungen bezüglich Art und Ausmaß des Pfortaderblockes geeignet, Untersuchungen zur Dynamik der Entwicklung einer portalen Hypertension und deren Pathophysiologie durchzuführen.
