As rats get older, alcohol-dehydrogenase (ADH) activity in the liver increases.

Alow protein diet (5% protein) decreases ADH-activity markedly as compared to controls fed a standard diet; however, after prolonged feeding of a low protein diet, ADH-activity returns to normal values.

A nearlyfree of protein diet (0.5%) results in a rapid fall of ADH-activity in the liver (about 15% of control values within 3 weeks). Under these conditions, ethanol oxidation is extremely slow as against blood ethanol concentration following a single oral dose of alcohol.

Ethanol administration brings about an initial increase of ADH-activity in both, rats fed the standard and the low protein diet. After ethanol is administered over a period of 3 weeks or more to animals maintained on the standard diet, no difference in ADH-activity is observed as compared to control animals.

Assuming that ethanol has a direct toxic effect on the liver cell, the decrease of ADH-activity might be a possible explanation for the unfavourable effect of protein deficiency in liver damage caused by alcohol.

Die Aktivität der ADH in der Rattenleber steigt mit zunehmendem Alter an.

Proteinarme Kost (5% Protein) senkt die ADH-Aktivität gegenüber normal ernährten Tieren deutlich. Nach längerer Gabe der proteinarmen Kost (6 Monate) gleichen sich die Werte denen der Kontrollen jedoch wieder an.

Fast eiweißfreie Kost (0,5% Protein) führt zu einem raschen Abfall der ADHAktivität in der Leber. Nach 3 Wochen liegt sie nur noch bei 15% der Ausgangsaktivität. Unter diesen Bedingungen ist der Alkoholabbau, gemessen an der Abnahme des Blutalkoholspiegels nach Alkoholzufuhr, extrem verlangsamt.

Unter äthanolzufuhr steigt die ADH-Aktivität bei normal und eiweißarm ernährten Ratten initial deutlich an. Nach einer Alkoholzufuhr über 3 Wochen und länger besteht kein Unterschied zu den Kontrollen mehr.

Bei der Annahme einer direkt toxischen Wirkung von Alkohol auf die Leberzellen ist durch die Verminderung der ADH-Aktivität eine mögliche Erklärung für die ungünstige Wirkung von Eiweißmangel bei Alkoholschäden der Leber gegeben.
