Zusammenfassung
Nach einem kurzen überblick über die wichtigsten Hypothesen über den Mechanismus der diabetogenen Wirkung von Alloxan wird in eigenen Untersuchungen die Frage geprüft, ob Alloxan etwa durch Verdrängung der Pyrimidinbasen Cytosin, Thymin und Uracil die Biosynthese von Nucleinsäuren beeinträchtigt. Dazu wurden diese Substanzen sowie Orotsäure, Ureidobernsteinsäure und (vergleichsweise) Barbitursäure in viermal so hoher Konzentration wie Alloxan weiÜen Ratten 5 min vor der Alloxanapplikation intraperitoneal injiziert. Unmittelbar vor der Injektion, dann nach 3 Std und nach 4 Tagen wurde der Blutzucker, schlieÜlich die Serumharnsäure und nach dem Töten der Tiere das Leberglykogen bestimmt. Lediglich Barbitursäure und Orotsäure konnten die Alloxanwirkung aufheben.
Summary
After a short survey on the most important hypotheses about the mechanism of the diabetogenic effect of alloxan it is the question experimentally examined, whether alloxan influences the biosynthesis of nucleic acids by competition of the pyrimidine bases cytosine, thymine and uracil. For this purpose these substances as well as orotic acid, ureido-succinic acid and (for comparison) barbituric acid were intraperitoneally injected to albino rats in a concentration fourth times as high as that of alloxan 5 min before the application of the latter. Immediately before the injection, after 3 hours and after 4 days blood sugar, serum uric acid and liver glycogen (after killing the animals) were determined. The effect of alloxan was nullified only by barbituric acid and by orotic acid.
