Zusammenfassung
In vergleichenden morphologischen und funktionellen Untersuchungen wurde der Einfluß von Glyoxal auf die Regulation des Kohlenhydrathaushaltes der Ratte geprüft. Glyoxal führt bei intravenöser Applikation zu einer zeitlich begrenzten Hypoglykämie. Wie in Versuchen an pankreatektomierten Ratten gezeigt wird, ist die Auslösung einer Hypoglykämie an das Vorhandensein des Pankreas gebunden. Es wird weiterhin gezeigt, daß Glyoxal ohne Zwischenschaltung anderer Regulationsmechanismen primär an den Langerhansschen Inseln angreift. Morphologisch dokumentiert sich der blutzuckerspezifische Glyoxaleffekt vornehmlich in einer Schädigung der B-Zellen. Die Kombination einer Hypoglykämie mit morphologischen Inselzellveränderungen im Sinne eines Diabetes mellitus wird erörtert.
Summary
The influence of glyoxal on the regulation of carbohydrate metabolism in the albino-rat was investigated by comparing morphological and functional findings under different conditions. Intravenously injected glyoxal causes a transitory hypoglycaemia. This phenomenon cannot be reproduced in pancreatectomized animals. Furthermore, glyoxal is shown to act primarily on the islets ofLangerhans without mediation by other regulatory mechanisms of carbohydrate metabolism. The histological alterations equivalent to the specific hypoglycaemic glyoxal effect chiefly consist of lesions of the B-cells of the islets ofLangerhans varying from degranulation and toxic edema to extensive necrosis. The combination of hypoglycaemia and B-cell lesions as sometimes found in diabetes mellitus is discussed.
