Zusammenfassung
Mittels horizontaler Stärkegelelektrophorese wurden im Hämolysat von je 40 Erwachsenen und Neugeborenen 2 Hexokinase-Isoenzyme nachgewiesen, die den Typen I und III aus Ratten- und Menschenleber entsprechen. Die gesteigerte Hexokinaseaktivität in Neugeborenenerythrocyten ist auf eine Aktivitätssteigerung des Isoenzyms I zurückzuführen, während bei hyperregeneratorischen Anämien (β-Thalassämie, kongenitale nichtsphärocytäre hämolytische Anämie unbekannter Ursache) die Aktivität beider Isoenzyme erhöht ist. Bei einem Fall von Fanconi-Anämie wurde ein relativer Hexokinasemangel ohne Verschiebung des Isoenzymmusters nachgewiesen. Eine Koppelung zwischen der Bildung der Hexokinase-Isoenzyme und der Bildung derγ-Ketten des Hämoglobins F ist auf Grund der Untersuchungen bei Neugeborenen, beiβ-Thalassämie und bei α-Thalassämie wenig wahrscheinlich.
Summary
Using horizontal high-voltage starch gel electrophoresis for the hemolysates of 40 adults and 40 newborn infants, two hexokinase isoenzyms could be demonstrated. Probably they correspond to the isoenzymes type I and III from rat and human liver. The increased hexokinase activity of newborn erythrocytes is due to an increase of the isoenzyme I activity, whereas in hyperregenerative hemolytic anemias (β-thalassemia, congenital nonspherocytic hemolytic anemia of unknown cause) the activity of both isoenzymes is increased. In one case of Fanconianemia a relativ deficiency of hexokinase together with a normal isoenzyme pattern could be demonstrated. There is no evidence for a connection between the formation ofγ-chains of hemoglobin F and the formation of hexokinase isoenzymes.
