Eine einfache Methode zur Tryptophan-Bestimmung in Proteinen

Summary

In alteration of a procedure of Scoffoneet al. a simplified method for quantitative determination of tryptophan residues in proteins is described. The modification of the tryptophan residues is carried out with 2-nitrophenylsulfenylchlorid and the change in the absorption at 365 nm is measured. Only 0.5–3 mg of proteins are required for this procedure. The absence of troubling SH-groups is checked by the specific method of Grassetti and Murray.

Zusammenfassung

In Abänderung eines Verfahrens nach Scoffoneet al. wird eine einfache Methode zur quantitativen Bestimmung von Tryptophan-Resten in Proteinen beschrieben. Die Modifizierung der Tryptophan-Reste erfolgt mit 2-Nitrophenylsulfenylchlorid und die dadurch bewirkte Extinktionsänderung wird bei 365 nm gemessen. Nur 0,5–3,0 mg Protein sind zur Durchführung des Tests erforderlich. Die Abwesenheit störender SH-Gruppen wird durch eine spezifische Methode nach Grassetti und Murray überprüft.
