Zusammenfassung
1. Der 2,3-Diphosphoglyceratgehalt beträgt in Erwachsenenerythrocyten 36,2µMole/1011 Zellen und in Neugeborenenerythrocyten 44,3 µMole/1011 Zellen. Der Unterschied ist nicht signifikant.
2. Während der Inkubation bei 36° C in glucosehaltigem Medium fällt in Neugeborenenerythrocyten der 2,3-Diphosphoglyceratgehalt innerhalb von 3 Std sechsmal schneller ab als in Erwachsenenerythrocyten. Dieser Unterschied ist statistisch gesichert.
3. Die Aktivitäten des die 2,3-Diphosphoglycerat-Bildung katalysierenden Enzyms 2,3-Diphosphoglyceratmutase und die Aktivitäten des den 2,3-Diphosphoglyceratabbau katalysierenden Enzyms 2,3-Diphosphoglyceratphosphatase unterscheiden sich in Erwachsenen-und Neugeborenenerythrocyten nicht. Unter optimalen Versuchsbedingungen (Substratüberschuß) ist die 2,3-Diphosphoglyceratbildung in Neugeborenenerythrocyten nicht vermindert.
4. Da die Durchflußrate der Glykolyse in Neugeborenenerythrocyten erhöht ist, wird angenommen, daß der starke Abfall des 2,3-Diphosphoglyceratgehaltes in Neugeborenenerythrocyten bei der Inkubation durch eine verminderte 2,3-Diphosphoglyceratbildung zustande kommt. Bei erhöhter Glykolyse kommt es deswegen zu einer Abnahme der 2,3-Diphosphoglyceratbildung, weil das für die 2,3-Diphosphoglyceratbildung notwendige Substrat 1,3-Diphosphoglycerat in erhöhtem Maße an der Phosphoglyceratmutase umgesetzt wird, so daß der 2,3-Diphosphoglyceratmutase das Substrat entzogen wird.
Summary
1. The erythrocytes of human adults have 2,3-diphosphoglycerate levels of 36 µmoles per 1011 cells, whereas the erythrocytes of newborn infants have levels of 44,3 µmoles per 1011 cells. The difference is not significant.
2. During incubation of red cells at 36° C in a medium containing glucose in erythrocytes of newborn infants the decrease of the 2,3-diphosphoglycerate levels is six times greater than in erythrocytes of adults.
3. The activities of the 2,3-diphosphoglycerate forming enzyme 2,3-diphosphoglycerate mutase and the activities of the 2,3-diphosphoglycerate hydrolyzing enzyme 2,3-diphosphoglycerate phosphatase show no difference in erythrocytes of adults and newborn infants. Under optimal experimental conditions (substrate in excess) the formation of 2,3-diphosphoglycerate is not diminished in erythrocytes of newborn infants.
4. Because of the high glycolytic rate in the erythrocytes of newborn infants it is suggested that the greater decrease of the 2,3-diphosphoglycerate levels during incubation in the erythrocytes of newborn infants is caused by a diminished formation of 2,3-diphosphoglycerate. This may be explained by the fact that of the two enzymes competing for the substrate 1,3-diphosphoglycerate, phosphoglycerate kinase and 2,3-diphosphoglyceratemutase, the first is favoured at a higher glycolytic rate, thus diminishing the formation of 2,3-diphosphoglycerate.
