Zusammenfassung
Es wird über ein einfaches, schnell durchzuführendes Verfahren zur Bestimmung der Aktivität der Pseudocholinesterase im Serum berichtet. Das Verfahren kann als Makro- oder Mikromethode angewandt werden und ist gut reproduzierbar. Es verwendet als Substrat Butyrylthiocholinjodid, das von PsChE umgesetzt wird. Das freigesetzte Thicholin reduziert 5,5-Dithiobis-2-nitrobenzoat zu gelb gefärbtem 5-Mercapto-2-nitrobenzoat; die Geschwindigkeit der Farbbildung (als kinetisches Verfahren angewandt) ermöglicht die Ermittlung der PsChE-Aktivität. Die Bestimmung wird durch die Acetylcholinesterase der Erythrocyten bei Hämolyse nicht gestört.
Summary
A method is described for the determination of pseudocholine esterase activity in serum. The test is simple, not time consuming, well reproducible and applicable to macro- and microscale as well.
The substrate butyrylthiocholineiodide reacts with pseudocholine esterase to yield thiocholine, which is able to reduce 5,5-dithiobis-2-nitrobenzoate to 5-mercapto-2-nitrobenzoate, recognizable by its yellow colour. The enzyme activity can be calculated from the velocity of colour production.
Acetylcholine esterase from erythrocytes (in case of haemolysis) does not interfere with the estimation of pseudocholine esterase activity.
