Zusammenfassung
Folsäure aktiviert und überträgt im C1-Stoffwechsel Formaldehyd und Ameisensäure. Unter der Vorstellung, diese toxischen Metabolite des Methanols beschleunigt zu eliminieren, erhalten methanolvergiftete Hunde (2 g/kg Methanol intravenös) Folsäure in einer Gesamtdosis von 30 mg/kg, verteilt auf 3 Tage nach der Vergiftung. Es gelingt so, die Methanolelimination zu beschleunigen und das Auftreten von Ameisensäure im Plasma und Harn fast vollständig zu unterdrücken. Bei Folsäuremangel, erzeugt durch Amethopterin (1,5 mg/kg per os 3 Tage vor Methanol), steigen dagegen die Ameisensäurewerte im Plasma auf mehr als das Doppelte, die Gesamtausscheidung im Harn auf mehr als das Fünffache an.
Summary
Folic acid affords the activation and the transfer of the C1-units, formaldehyde and formic acid, known as metabolites of methanol. An acceleration of the elimination of these toxic metabolites is achieved in dogs poisoned with methanol (2 g/kg i.v.) by intravenous doses of folic acid, 30 mg/kg divided into 3 daily dosages following the administration of methanol. This treatment results in a decrease of the actual methanol concentration and in an almost complete suppression of the appearance of formic acid in plasma, as well as of its excretion with urine. On the other hand, in the state of folic acid deficiency, induced by 1.5 mg/kg amethopterine 3 days before the methanol administration, the formic acid plasma levels amount to more than twice the control values, and the total urinary output increases to fivefold the amount found in methanol poisoned dogs without amethopterine.
