Zusammenfassung
Ein neuer, einfacher, durch Coulter-Zählung halbautomatisierter In-vitro-Test, der auf unterschiedlicher Glashaftung isolierter und inkubierter Blutleukozyten (Lymphozyten und Monozyten) nach Antigenberührung beruht, wird beschrieben. 15 Kinder mit Neuroblastom, Nephroblastom, leukämischem Lympho-(Retikulo-)sarkom, Morbus Hodgkin und Hepatokarzinom wurden damit untersucht. Wasserlösliche, durch Ultrazentrifugierung gewonnene Tumorextrakte dienten als Antigene. Bei fast allen Patienten, besonders solchen mit Neuroblastom, konnten tumorspezifisch sensibilisierte Lymphozyten durch Testung mit autologen sowie mit homologen Tumorextrakten statistisch signifikant nachgewiesen werden. Die antigeninduzierte Leukozytenhaftungshemmung konnte tumorspezifisch durch Zusatz von Serum von Kindern mit wachsenden, generalisierten Tumoren meist völlig aufgehoben werden. Diese „blockierenden Serumfaktoren“ scheinen hinsichtlich Verlauf und Prognose bei Krebspatienten eine wichtige Rolle zu spielen.
Summary
A new, simple, half-automated (Coulter-Counter) in-vitro test is described which is based upon differing glass adherence of isolated and incubated blood leukocytes (lymphocytes + monocytes) after antigen contact. 15 children with neuroblastoma, nephroblastoma, leukemic lypho(reticulo)sarcoma, Hodgkin's disease and hepatocarcinoma were examined. Watersoluble tumor extracts, obtained by ultracentrifugation, served as antigens. In most of the patients, especially those with neuroblastoma, tumorspecific sensitized lymphocytes could be demonstrated by testing with autologous as well as with homologous tumor extracts. The antigen-induced leukocyte adherence inhibition could be blocked specifically and almost completely by serum of patients with generalized, growing tumors. These “serum blocking factors” seem to play an important role as to course and prognosis in patients with malignant diseases.
