Zusammenfassung
Nach intraduodenaler Application von gereinigtem125J-markiertem Human-Trypsin wurden beim Menschen nach 15–30 min 4–6% der Markierung im Blutplasmavolumen gefunden. Die Radioaktivität im Blutplasma befand sich bei der Auftrennung durch Dextrangelfiltration in den gleichen Fraktionen, die auch nach Inkubation von Human-Trypsin (-125J) mit Humanserum abgetrennt wurden. Im aus dem Pankreasgang aspirierten Pankreassaft wurde schon nach 20 min etwa 1% der verabfolgten Trypsin(-125J)-Dosis gefunden. Die Ergebnisse stützen die Annahme der Existenz eines enteropankreatischen Kreislaufs auch beim Menschen und erklären z.T. auch die gemessenen niedrigen bis nicht mehr nachweisbaren Mengen an immunoreaktivem Serum Trypsin bei Patienten mit exkretorischer Pankreasinsuffizienz.
Summary
Following the intraduodenal installation of purified125I-labeled human trypsin up to about 4–6% of the label was measured after 15–30 min in blood plasma and found to separate in a dextrangel filtration system similar to purified human trypsin (-125I) after incubation with human serum. About 1% of the installed trypsin-(-125I)-dose was found already after 20 min in 100 ml of aspirated pancreatic secretion and later on also in the duodenal content. The results support the concept of the existence of an enteropancreatic circulation of trypsin also in man and explain in part the low to non-detectable levels of immunoreactive serum trypsin observed in patients with exocrine pancreatic insufficiency.
