Zusammenfassung
Unter der Infusion von DOPA wird an sechs Hypertonikern das Blutdruckverhalten vor und während der Verabreichung von α-methyl-DOPA (Aldomet) untersucht. Dabei zeigt sich, daß nach einer mindestens dreitägigen Aldomet-Medikation eine vorübergehende, signifikante Blutdrucksteigerung eintritt, während dieselbe Infusion am unbehandelten Patienten den Blutdruck nicht verändert.
Das Hypotensivum α-methyl-DOPA hemmt zwar diel-aromatische Aminosäuredecarboxylase, besitzt aber daneben die Eigenschaft, den Gewebsgehalt an Noradrenalin langdauernd zu senken. Diese letztgenannte Wirkung dient möglicherweise zur Interpretation des beobachteten Blutdruckverhaltens, indem ein kompetitiver Mechanismus zwischen dem α-methylierten und dem endogenen Noradrenalin angenommen wird. Bekanntlich entstehen im Organismus diese beiden Amine in analoger Weise aus α-methyl-DOPA bzw. DOPA. Die ursprünglich allein bekannte Decarboxylasehemmung durch α-methyl-DOPA scheint demgegenüber in den Hintergrund zu treten.
Summary
Changes in blood pressure during an infusion of DOPA have been studied in 6 hypertensive patients before and under medication of α-methyl-DOPA (Aldomet). While in untreated patients no alteration in blood pressure was observed, a significant rise of blood pressure was found in hypertensives after a period of Aldomet administration of at least 3 days.
α-methyl-DOPA inhibits thel-aromatic amino acid decarboxylase. Moreover it lowers the content of norepinephrine in tissues, an effect which might explain our findings on the basis of a possible competitive mechanism between the α-methylated and the endogenous norepinephrine.
