Zusammenfassung
Bei 15 normalen Hunden wurden den Herzen (linker Ventrikel) eine Kontrollprobe und, nach Unterbinden des Kreislaufs (Ischämie), sieben weitere Proben in 3 min-Abständen entnommen und durch „Frierstopp“ fixiert. Der Gehalt der Muskelproben an Phosphokreatin, ATP, ADP, AMP und anorganischem Phosphat wurde analysiert.
Die Gewebsgehalte für ATP, ADP und CP wurden in ihrer gegenseitigen Abhängigkeit entsprechend dem Kreatinkinase-Gleichgewicht untersucht. Unter Berücksichtigung der pH-Abhängigkeit des Kreatinkinase-Gleichgewichts13, 14 wurde der ischämie-bedingte pH-Abfall in der Zelle berechnet und die funktionelle Bedeutung der Milchsäure-Anhäufung für die Phosphatverschiebung vom Phosphokreatin zum ADP bestätigt.
Summary
A thoracotomy was performed on 15 mongrel dogs who were anesthesized with pentothal-succinylcholine anesthesia. Left ventricular biopsy specimens (approximatly 500 mg) were immediately grasped by aluminum tongs precooled in liquid air. The ascending aorta and venae cavae were then severed and at 3 min intervals seven additional biopsies were taken by the same technic. The muscle samples were analyzed for CP, ATP, ADP, AMP and inorganic phosphate. Other cofactors and substrates of the Embden-Meyerhof-pathway were analyzed simultaneously and these data have been published in an earlier paper1.
The time course of the ATP, ADP and CP content of heart muscle under anoxic conditions was investigated with special regard for the creatine kinase equilibrium. After accounting for the pH-dependency of the creatine kinase equilibrium the decline of the pH during the anoxic period was calculated. The presence of the lactic acid accummulation for the disposal of the creatine phosphate, as assumed by other authors, was confirmed.
