Summary
Investigations on the stimulation of glutathione reductase of human erythrocytes by riboflavin and FAD showed the following results:

In glutathione reductase deficiency, the enzyme activity in the optical test was normalized by addition of FAD. The pH optimum shifted from pH 6.42±0.13 to 6.76±0.06. Glucose-6-phosphate dehydrogenase and methemoglobin reductase showed no changes of enzyme activity.

Patients with glutathione reductase deficiency demonstrated normal enzyme activity and Heinz body formation after administration of riboflavin. GSH-content, -stability, and -reduction of glutathione reductase-deficient erythrocytes were not influenced. The methemoglobin reductase was not stimulated by riboflavin.

Patients with hematological diseases and glutathione reductase deficiency demonstrated no improvement in spite of normalized enzyme activity.
Zusammenfassung
Versuche zur Aktivierung der Glutathionreductase menschlicher Erythrocyten mit Riboflavin und FAD zeigten folgende Ergebnisse:

Bei Glutathionreductase-Mangel wurde die Enzymaktivität im enzymatischen Test durch FAD-Zusatz normalisiert. Das pH-Optimum der Enzymreaktion wurde von 6,42±0,13 nach 6,76±0,06 verschoben. Die Glucose-6-phosphat-Dehydrogenase und das FAD-haltige Enzym NADH-abhängige Methämoglobinreductase zeigten keine Aktivitätsänderung.

Nach oraler Gabe von Riboflavin wurden bei Glutathionreductase-Mangel eine Normalisierung der Enzymaktivität und des Heinzkörpertests sowie ein normales pH-Optimum der Enzymreaktion beobachtet. GSH-Gehalt, -Stabilität und-Reduktionsgeschwindigkeit der Erythrocyten zeigten keine Veränderungen. Die NADH-abhängige Methämoglobinreductase wurde durch FAD nicht aktiviert.

Bei Patienten mit hämatologischer Erkrankung und Glutathionreductase-Mangel trat trotz Normalisierung der Enzymaktivität keine Änderung des klinischen Krankheitsbildes auf.
