Zusammenfassung
In den Meristemen und sich differenzierenden Geweben der Wurzel und des Blattes vonAllium carinatum wurden an denselben Zellkernen Chromatinstruktur, Volumen und DNS-Gehalt parallel untersucht. Sowohl während der mitotischen, als auch während der endomitotischen Interphase zeigt sich ein Unterschied in der Struktur zwischen Kernen im Stadium G1 und solchen in G2: Die in G1 locker verteilten, kleinen Euchromomeren werden größer und erfüllen in G2 regelmäßiger verteilt und dichter gelagert den Kernraum; die kompakten Chromozentren wachsen ebenfalls heran und verschmelzen zusätzlich zu Sammelbildungen, so daß ihre Anzahl in G2 verringert erscheint. Die Strukturänderungen während der S-Periode selbst sind schwieriger zu beurteilen, jedoch bleiben die Chromozentren sicher während der DNS-Replikation kompakt.
Im Zuge der mitotischen Prophase bzw. der Endomitose kommt es zu einer völligen Angleichung der euchromatischen und heterochromatischen Chromatinelemente im Zerstäubungsstadium (Z); seine Bedeutung liegt offenbar in der Trennung der Chromatiden. da unmittelbar nachher die für G1 charakteristische feinkörnige Struktur wiederhergestellt wird und auch die Chromozentren wieder verkleinert, aber vermehrt hervortreten (Stadium der Neu-Organisation, NO).
In der Mitose erscheinen im Anschluß daran, während der frühen Spiralprophase, Eu- und Heterochromatin vorerst wieder aneinander angeglichen (Stadium SpE), später kommt es aber infolge des asynchronen Spiralisationsverhaltens der heterochromatischen Abschnitte der Chromosomen zu einer neuerlichen Differenzierung (Stadium SpH), und erst in einem mittleren Prophasestadium sind die Chromosomen endgültig einheitlich stark spiralisiert (SpS).
Das Kernwachstum erfolgt sowohl im mitotischen als auch im endomitotischen Kernzyklus in drei Schühen: Der erste findet unmittelbar nach der mitotischen bzw. endomitotischen Telophase statt, der zweite nach der DNS-Synthese, der dritte kurz vor und während der mitotischen Prophase bzw. der Endomitose. Die Volumenzunahme zwischen G1 und G2 sowie zwischen G1 und der Zerstäubung sind im mitotischen Kernzyklus größer als im endomitotischen.
