Zusammenfassung
Primäre stationäre Affennierengewebekulturen (Cerc. aeth.) wurden 24 Stunden lang vor ihrer Infektion mit SV 40 mit14C-Thymidin markiert. 24 und 48 Stunden nach Infektion wurde jeweils ein Teilder Kulturen mit3H-Desoxyadenosin und 5-Jod-2′-desoxyuridin versetzt; 24 Stunden danach wurde die DNS dieser Kulturen extrahiert und in CsCl-Dichtegradienten zentrifugiert. Hierbei stellte sich der durch JUDR-Einbau schwerer gewordene DNS-Anteil als zweiter Gipfel dar. Dieser Gipfel besaß sowohl3H- als auch14C-Aktivität. Die schwere DNS aus infizierten Kulturen wies mehr14C-Aktivität auf als die schwere DNS aus nicht infizierten Kulturen. Da die14C-Aktivität ausschließlich vor der Infektion in die Zell-DNS eingebaut worden war, folgt daraus, daß die Infektion stationärer Affennierengewebekulturen mit SV 40 die Synthese zellulärer DNS induziert. Wurden die Kulturen vor der Infektion UV-bestrahlt oder vor und nach der Infektion mit Mitomycin C behandelt, so wurde diese Induktion gehemmt bzw. unterdrückt, ohne daß die Vermehrung von infektiösem Virus reduziert wurde. Die durch den SV40-Infekt hervorgerufene Aktivitätssteigerung der Desoxycytidin-Kinase, Thymidylat-Synthetase, Thymidin-Kinase und DNS-Polymerase wurde durch die UV-Bestrahlung oder Mitomycin-Behandlung im wesentlichen nicht beeinflußt.
Summary
Primary monkey kidney cell cultures (Cerc. aeth.) in stationary growth phase were labeled with14C-thymidine for 24 hours prior to infection with SV40.3H-deoxyadenosine and 5-iodo-2′-deoxyuridine were added to some of the cultures 24 and 48 hours after infection; 24 hours later the DNA was extracted from these cultures and centrifuged in a CsCl density gradient. The portion of DNA which had become heavier due to incorporation of IUDR could be seen as a second peak in the sedimentation profile. This peak contained14C as well as3H activity. More14C activity had been incorporated into the heavier DNA of the infected cultures than into the heavier DNA of the noninfected cultures. Since the cellular DNA only had been labeled prior to infection, it was concluded, that the SV40-infection of stationary monkey kidney cell cultures induces the synthesis of cellular DNA. This induction was inhibited by treatment with UV or mitomycin C prior respectively prior and after infection. However, synthesis of infectious virus was not reduced. Under these conditions the stimulation of the activity of the enzymes deoxycytidine-kinase, thymidylate-synthetase, thymidine-kinase, and DNA-polymerase was not markedly influenced.
