Zusammenfassung
Für toxikologisch-chemische Untersuchungen sind einfache, rasche und sichere Methoden der Trennung komplizierter Substanzgemische unentbehrlich. Neben den bekannten Methoden wie Papierchromatographie und fraktionierte Sublimation kann die auf dem Prinzip der „offenen Säule“ beruhende Dünnschichtchromatographie erfolgreich für viele Substanztrennungen verwendet werden. Diese adsorptionschromato-graphische Methode an Kieselgelschichten erlaubt in kürzester Zeit eine Trennung der verschiedenen organischen Gifte und überdies die Entwicklung der Chromatogramme auch mit aggressiven Sprühreagenzien. Die Technik der Anfertigung geeigneter Schichten und die praktische Anwendung der Methode für toxikologische Probleme wird ausführlich geschildert.
Summary
Simple, rapid and reliable methods for separating complicated mixtures are indispensable to toxicological-chemical studies. Besides the familiar procedures such as paper chromatography and fractional sublimation, many separation problems can be successfully solved by thin layer chromatography which is based on the principle of “the open column”. This adsorption Chromatographic method on layers of silica gel permits a rapid separation of various organic poisons and in addition the development of the chromatogram by means of aggressive spray reagents. A detailed description is given of the technique of preparing suitable layers and of the practical application of the method to toxicological problems.
Résumé
Il est indispensable de disposer de méthodes de séparation de mélanges complexes simples, rapides, sûres pour les recherches de toxicologie chimique. A côté des méthodes connues telles que la Chromatographie sur papier et la sublimation fractionnée il est possible d'employer pour de nombreuses séparations de substances le principe de la «colonne ouverte» qui repose sur la Chromatographie en couches minces. Cette méthode d'absorbtion chromatographique sur des couches de gel de silice permet de séparer dans un temps très court les différents poisons organiques et de développer ensuite le chromatogramme à l'aide de réactifs pulvérisés également agressifs. La technique de préparation de couches adsorbantes appropriées et la mise en œuvre pratique de la méthode pour la résolution de problèmes de toxicologie sont décrites en détail.
