Abstract
Endoglucanase (carboxymethylcellulase) production byBacillus thermoalkalophilus was both inducible and repressible. The enzyme production by free and immobilized cells was studied. Of the four entrapment matrices that were tried, polyacrylamide was the best at retaining cells at 60°C. The rate of CMCase production by immobilized cells was low initially, but increased and siabilized by day 7. Change in the metabolic behaviour of the immobilized cells was envisaged due to low oxygen consumption.
Résumé
La production d'endoglucanase (carboxymethylcellulase) parBacillus thermoalkalophilus est à la fois inductible et répressible. On a étudié la production d'enzyme par des cellules libres et immobilisées. Des quatre matrices de piégeage essayées, le polyacrylamide s'est avéré le meilleur pour retenir les cellules à 60°C. La vitesse de production de la CMCase par les cellules immobilisées était faible au début, mais s'est accrue et stabilisée au septième jour. On envisage un changement de comportement métabolique, dû à la faible consommation du substrat.
