Summary
This review paper deals with the physico-chemical state of the water and the salts in the living cell. Measurements of the dielectric constant of protein solutions, cells and tissue show that only a small amount of the cell water — 10 to 20% — is bound to protein molecules. The rest behaves like normal water. The same result has been obtained by calorimetric measurements of the heat of melting and of the partial specific heat in protein solutions and in cells. From the results of other experiments on the determination of the specific electrical conductivity of the cytoplasm in several cell types the conclusion may be drawn, that practically all of the intracellular ions are solved in the cell water and show an electrical mobility essentially not different from the value in an electrolyte solution. Furthermore, measurements of the activity coefficient of KCl and NaCl in protein solutions of high concentration and in soluble fractions of cytoplasm have been described. The experimental results show with little doubts that within the physiological range of the experimental conditions the activity coefficient of salts in the cytoplasma — aside from ion binding — is not altered seriously by the presence of large amounts of proteins, compared to a simple salt solution. An explanation of this strange result will be presented.
Zusammenfassung
In der vorliegenden Übersichtsarbeit werden Versuchsergebnisse besprochen, die eine Aussage über den physikalisch-chemischen Zustand des Wassers und der Salze in der lebenden Zelle erlauben. Messungen der Dielektrizitätskonstante in Proteinlösungen, Zellen und Geweben führen zu dem Schluß, daß nur ein kleiner Teil des Zellwassers, 10 bis 20%, an Proteine gebunden ist und daß der größte Teil frei ist. Zum gleichen Ergebnis führen kalorische Messungen der Schmelzwärme und der partiellen spezifischen Wärme de Wassers. Auch die Analyse der spezifischen elektrischen Leitfähigkeit im Zytoplasma verschiedener Zellen kann man mit der Annahme erklären, daß die intrazellulären Ionen im Zellwasser gelöst sind und sich unter dem Einfluß des elektrischen Feldes im Wasserraum des Zytoplasmas wie in einer Elektrolytlösung bewegen. Messungen des thermodynamischen Aktivitätskoeffizienten von KCl und NaCl in hochkonzentrierten Proteinlösungen und in löslichen Zytoplasmafraktionen zeigen unzweideutig, daß unter physiologischen Bedingungen die Aktivität von Salzen im Zytoplasma — abgesehen von spezifischen Ionenbindungen — durch die Anwesenheit von Protein nur sehr wenig reduziert wird. Diese geringe Reduktion wird erklärt.
