Linear pharmacokinetic models: Geometric construction to determine transfer and elimination rate constants

Abstract

It is shown that, in linear pharmacokinetics, the blood concentration-time curves determine the model to a certain extent, but not completely. Intervals of indetermination for the parameters are calculated and represented by a simple graph. Each additional measurement (of unchanged drug in urine, of metabolites in blood or in urine) will help to reduce these intervals. A unique model can be elaborated only in certain cases, or with additional hypotheses. Utilizing data from the literature, numerical applications are detailed.
