Summary
A method for sample preparation has been modified and developed for the determination of hydroxycinnamic acid derivatives. After careful extraction the extract is fractionated by column chromatography on polyamide. Two fractions are obtained. The methanolic fraction contains the derivatives of hydroxycinnamic acids with glucose (ester and glucosides), and the methanol/formic acid extract of the polyamide column contains the quinic, tartaric and malic acid esters. This method was tested and confirmed with reference samples as well as with vegetable samples. Additionally this method effects a sample clean up.
The samples were analysed by HPLC using a methanol gradient on a reversed phase system.
In addition to the differentBrassicas we investigated summer-squash, globe artichoke, leek, corn salad, rhubarb, scorzonera and carrot.
The compounds analysed were found in greatly varying amounts in vegetable depending on the part of the plant analysed.
Zusammenfassung
Zur Bestimmung von Hydroxyzimtsäure-Derivaten in Pflanzenextrakten (hier: Gemüse) wurde eine Methode der Probenaufarbeitung modifiziert und weiterentwickelt. Nach schonender Extraktion des Probengutes erfolgt eine Fraktionierung des Extraktes durch Säulenchromatographie an Polyamid. Dadurch werden zwei Fraktionen erhalten, die im Methanol-Extrakt die Glucoseverbindungen (Glucoseester und Glucoside) und in Methanol/ Ameisensäure-Extrakt die Gesamtheit der China-, Wein- und Äpfelsäureester der Hydroxyzimtsäuren enthalten. Die Fraktionierung wurde quantitativ nicht nur an Reinsubstanzen, sondern auch an Gemuseproben überprüft und bestätigt. Gleichzeitig bewirkt die Fraktionierung eine Proben-Reinigung
Die Gemüseproben werden hochleistungsflüssig-chromatographisch mit einem Methanol-Gradienten an Phasen-Umkehr-Trennsaulen untersucht und die Hydroxyzimtsaure-Derivate quantitativ erfaßt.
Neben den Kohlarten werden noch Zucchini, Artischocken, Porree, Feldsalat, Rhabarber, Schwarzwurzeln und Möhren untersucht. Die analysierten Verbindungen kommen im Gemuse, abhängig vom geprüften Pflanzenteil, in stark wechselnden Gehalten vor.
