Zusammenfassung
Mit Hilfe der direkten und indirekten Fluorescenzmethode nachCoons gelang es, Toxoplasmen in Milz, Lymphknoten und Leber von intraperitoneal infizierten MÄusen und Meerschweinchen spezifisch anzufÄrben. Am krÄftigsten leuchtete die ZelloberflÄche der Parasiten auf, die als Ort der Antigen-Antikörper-Reaktion anzusehen ist. WÄhrend die Toxoplasmen im akuten Stadium der Infektion intakt intraoder extracellulÄr nachweisbar waren, hatten sie im chronischen Stadium ihre scharfe Begrenzung verloren und waren nur noch an der für Toxoplasmen spezifischen Fluorescenz erkennbar, und zwar nur noch im Lebergewebe. Die StÄrke der Fluorescenz war abhÄngig von der Höhe des nachSabin-Feldman gemessenen Titers der zur FÄrbung benutzten Sera: Je höher der Titer, desto krÄftiger die Fluorescenz der Toxoplasmen. Ihre Darstellung mit dieser Methode zeigt, da\ dem Sabin-Feldman-Test bei der Antikörpermessung der Toxoplasmose eine spezifische Bedeutung zukommt.
Summary
Using the Coons fluorescein labelled antibody technic (direct and indirect methods), toxoplasma can be specifically stained in the spleen, lymph nodes, and liver of experimentally infected mice and guinea pigs. The outer cellmembran of the organisms fluoresces most intensely, indicating probably the site of antigen-antibody reaction. In the acute stages of infection the organisms are demonstrable intact intra- and extracellularly. In the chronic stages they lose the quality of giving a sharply demarcated staining and may be recognized only in the liver, and only by the presence of the specific fluorescence. The intensity of the fluorescence is dependent on the titer of the serum used for staining, as measured by the Sabin-Feldman test. The higher the titer the more brilliant the fluorescence. The demonstration of toxoplasma with the Coons technic confirms the opinion that the Sabin-Feldman test has specific significance in evaluating the titer of antibody against toxoplasma.
