Summary
(4-14C) Dehydroepiandrosterone (DHA) as oestrogen precursor has been incubated with microsomal fractions of human placentae from normal and pathological pregnancies at different ages of gestation, in order to evaluate the rate of conversion of the substrate to oestrogens. In the placentae from normal pregnancies the capability to convert C19- to C18-steroids was related to the age of gestation. The mature placenta metabolizes (4-14C) DHA to oestrogens (oestrone and oestradiol-17β) four times as fast as the placenta of midpregnancy. The concentration of protein in the placental microsomal fraction increased with the age of gestation. In our investigation it was found that the conversion of DHA to oestrogen (oestrone and oestradiol-17β) by placental microsomes was lower in the toxemic and diabetic placentae as compared to normal placentae. The relationship between placentar insufficiency and low capacity of theΔ 4-Δ 5-isomerase, 3β-hydroxysteroid-dehydrogenase and the aromatizing enzyme system will be discussed.
Zusammenfassung
(4-14C) Dehydroepiandrosteron (DHA) wurde als Oestrogenvorläufer mit der Mikrosomenfraktion von normalen und pathologischen Placenten verschiedener Gestationsalter (13.–40. Woche) inkubiert, um die Konversionsrate zu Oestrogenen (Oestron und Oestradiol-17β) zu messen. Mit zunehmendem Gestationsalter steigt die Umwandlungsrate von DHA zu Oestrogenen in der Mikrosomenfraktion normaler Placenten stark an (von 0–22,7%). Der Proteingehalt pro 1 ml Placenta-Mikrosomenfraktion nimmt ebenfalls im Verlaufe der Schwangerschaft zu. Die Placenten diabetischer Schwangerer ebenso wie diejenigen von Gestose-Patientinnen zeigen eine verminderte Umwandlungsrate von C19-zu C18-Steroiden. Der Proteingehalt der Placenta-Mikrosomenfraktion war bei allen untersuchten pathologischen Schwangerschaften niedriger als in den Normalfällen. Demnach besteht bei pathologischer Schwangerschaft mit Placentainsuffizienz eine quantitative Herabsetzung der Umwandlungsrate von DHA in Oestron und Oestradiol-17β. Die Zusammenhänge zwischen Placentainsuffizienz, mangelhafter steroidbiogenetischer Kapazität und einigen klinischen Fragen werden diskutiert.
