Summary
In dependence of various functional and developmental phases of the corpus luteum, luteal cells of the rat show changes in their ultrastructural appearance. Because mitochondria acquire a central position in cellular metabolic processes, changes in structure and size of these organelles allow conclusions on activity changes of the cell.
Supposition on resolved conclusions is, of course, an exact biochemical demonstration of such fluctuations in size of cell organelles in the electron microscopic picture. It was able by means of application of histometry into the ultrastructural area.
With ultrahistometry the smallest average diameter of lutein cell mitochondria was found during dioestrus, the longest in day 8 of pregnancy. In contrary to this phase a significant decrease of mitochondrial size during the last third of gestational period was particularly striking. Still more decreased was the average diameter of mitochondria in the corpus luteum lactationis. On the contrary a significant difference in size was missed between the mitochondria of lactating phase and oestrus phase.
Ultrahistometry by means of relatively quantitative representation of morphocinetic changes in cell organelles helps to inaugurate subtile insights into connections of function and structure in the cell. The combination of ultrahistometry with adequate mathematical and statistical methods of procedure and a strict observation of equal conditions of preparation have to be absolutely noticed.
Zusammenfassung
Die Luteinzellen der Batte zeigen in Abhängigkeit von verschiedenen Funktions- und Entwicklungsphasen des Corpus luteum typische Wandlungen im Erscheinungsbild ihrer Feinstruktur. Da die Mitochondrien im cellulären Stoffwechselgeschehen eine zentrale Stellung einnehmen, sind aus Veränderungen in Struktur und Größe dieser Organellen Rückschlüsse auf Aktivitätsveränderungen der Zelle erlaubt.
Voraussetzung für schlüssige Folgerungen ist allerdings eine exakte biometrische Objektivierung derartiger Größenschwankungen an Zellorganellen im elektronenmikroskopischen Bild. Sie gelang durch erstmalige Anwendung der Histometrie im ultrastrukturellen Bereich.
So wurde mit Hilfe derUltrahistometrie an den Luteinzellmitochondrien der kleinste mittlere Durchmesser im Diöstrus, der größte am 8. Tag der Gravidität nachgewiesen. Gegenüber diesem Stadium fiel ein signifikanter Rückgang der Mitochondriengröße im letzten Drittel der Tragzeit besonders auf. Noch kleiner wurde der mittlere Durchmesser der Mitochondrien des Lactationsgelbkörpers ermittelt. Zwischen der Lactationsphase und der Oestrusphase fehlte dagegen eine signifikante Größendifferenz der Mitochondrien.
Die Ultrahistometrie hilft durch relativ-quantitative Objektivierung morphokinetischer Veränderungen an Zellorganellen im elektronenmikroskopischen Bereich subtile Einblicke in Zusammenhänge zwischen Funktion und Morphe der Zelle eröffnen. Voraussetzung dafür ist allerdings ihre Kombination mit adäquater mathematisch-statistischer Methodik und die strikte Einhaltung gleichbleibender Präparationsbedingungen am Untersuchungsmaterial.
