Rapid determination of ergosterol and total minor sterols in the unsaponifiable of mould mycelium

Abstract

A new method for the determination of ergosterol and total minor sterols present in the unsaponifiable of the mycelium of Penicillium chrysogenum is described. The amount of total sterols can be found by means of the Liebermann-Burchard reaction, whilst the amount of ergosterol ist determined by fast, quantitative thin-layer chromatography on high-performance layers. The minor sterols result from the difference of both values. The 95% confidence limit is 5.3% (N=10).

Zusammenfassung

Zur Bestimmung von Ergosterin und der Gesamt-Nebensterine im Unverseifbaren des Mycels von Penicillium chrysogenum wird eine neue Methode beschrieben. Den Gehalt an Gesamtsterinen ermittelt man mit der Liebermann-Burchard-Reaktion, den an Ergosterin mit schneller, quantitativer Dünnschicht-Chromatographie an Hochleistungsschichten. Die Nebensterine ergeben sich aus der Differenz beider Werte. Der Standardfehler des Mittelwertes für den prozentualen Ergosterinanteil beträgt 5,3% (95% statistische Sicherheit,N = 10).
