Summary
Atropine is a competitive antagonist to acetylcholine (ACh) as well as to histamine. The affinity of atropine to the ACh receptor of the guinea pig ileum is about 5,000 times higher than to the histamine receptor of this organ. In order to obtain more information on the structure which may be responsible for this high specificity, the tropa acid part of the atropine molecule was altered systematically in 7 atropinelike analogues. The pharmacological activity of the compounds synthesized was tested on the guinea pig isolated ileum. After quantitative evaluation of the experimental results, a competitive antagonism to carbachol as well as to histamine could be demonstrated for all of the compounds synthesized. The affinity of the tropine esters to the ACh and to the histamine receptor of the guinea pig ileum was established. The alcoholic hydroxy group in a critical distance to the carbonyl group is responsible for the high specificity of atropine to the ACh receptor.
Zusammenfassung
Atropin ist ein kompetitiver Antagonist des Acetylcholin (ACh) wie auch des Histamins und hat zum ACh-Receptor des Meerschweinchen-Ileum eine 5000 fach höhere Affinität als zum Histamin-Receptor dieses Organs. Zur näheren Charakterisierung der für diese hohe Spezifität verantwortlichen Struktur änderten wir in 7 Atropin-Analogen die Tropasäure des Atropinmoleküls systematisch ab, ihre pharmakologische Wirkung wurde am isolierten Meerschweinchen-Ileum untersucht. Über eine quantitative Auswertung der Versuchsergebnisse konnte für alle Substanzen ein kompetitiver Antagonismus sowohl gegenüber Carbachol als auch gegenüber Histamin nachgewiesen werden. Die Affinität der untersuchten Tropinester zum ACh- und zum Histamin-Receptor des Meerschweinchen-Ileum wurde bestimmt. Für die hohe Spezifität des Atropin zum ACh-Receptor ist die alkoholische Hydroxygruppe in bestimmtem Abstand zur Carbonylgruppe verantwortlich.
