Summary
Some aminostilbene-derivatives were tested for carcinogenic activity in the rat by stomach-tube-feeding. The known carcinogen trans-4-dimethylaminostilbene (trans-DAS) produced earduct-tumors with high yields. Equimolar doses of 4-dimethylaminobibenzyl (DABB) and 4-dimethylaminotolane (DAT) were inactive in 18 months. With cis-4-dimethylaminostilbene (cis-DAS) no earduct-tumors were observed, but weak activity could not be excluded, because one Leukemia and one mammary-tumor occurred.
The chemically reactive derivatives trans-N-hydroxy-4-acetylaminostilbene (N-hydroxy-AAS) and trans-N-acetoxy-4-acetylaminostilbene (N-acetoxy-AAS) showed the organotropic activity of aminostilbenes in the feeding experiment and produced earduct-tumors in females. Males died from toxic effects but did not develop any tumors. N-acetoxy-4-acetylaminobibenzyl (N-acetoxy-AABB) was neither toxic nor carcinogenic.
Zusammenfassung
Einige Aminostilben-Derivate wurden auf krebserzeugende Wirksamkeit bei der Ratte geprüft nach Füttern mit der Schlundsonde.
Trans-4-Dimethylaminostilben (trans-DAS) erzeugte Plattenepithelcarcinome des äußeren Gehörgangs mit hoher Ausbeute. Äquimolare Dosen von 4-Dimethylaminobibenzyl (DABB) und 4-Dimethylaminotolan (DAT) waren in 18 Monaten inaktiv. Mit cis-4-Dimethylaminostilben (cis-DAS) wurden keine Gehörgangstumoren beobachtet. Schwache carcinogene Wirksamkeit dieser Verbindung kann wegen des Auftretens einer Leukämie und eines Mammacarcinoms nicht ausgeschlossen werden.
Nach Füttern der reaktionsfähigen Derivate trans-N-Hydroxy-4-acetylaminostilben (N-Hydroxy-AAS) und trans-N-Acetoxy-4-acetylaminostilben (N-Acetoxy-AAS) blieb die organotrope Wirkung der Stilbenamine erhalten und es entstanden Gehörgangstumoren bei den Weibchen. Die Männchen starben aufgrund toxischer Wirkungen, ohne Tumoren zu entwickeln. N-Acetoxy-acetylaminobibenzyl (N-Acetoxy-AABB) war weder toxisch noch carcinogen wirksam.
