Standardisierung Polyomavirus-induzierter Rattentumoren

Summary

The polyoma virus BB/T2 of Graffi was continuously cultivated for six years in embryonic mouse cell cultures. With several virus charges various sarcomas and carcinomas were induced in inbred Wistar rats by injecting the virus in newborn animals. The following five tumors were transplantated over years in continous lines: a liposarcoma, a rhabdomyosarcoma, a fibrosarcoma, an alveolar adenocarcinoma of salivary gland, and a solid carcinoma. These tumors grew in the recipient animals subcutaneously and developed at a constant rate and with constant histomorphology. From each tumor-line cell cultures were maintained which on reimplantation in young rats grew into solid tumors. Special investigations as DNA and RNA isolation and other analyses may be performed on these five tumors induced by the same virus.

Zusammenfassung

Das Polyomavirus BB/T2 von Graffi wurde seit 6 Jahren fortlaufend auf Mäuseembryonalzellkulturen gezüchtet. Mit einzelnen Viruschargen wurden durch Injektion in neugeborene Wistar-Ratten eines Inzuchtstammes verschiedene Sarkome und Carcinome induziert. Von diesen wurden folgende 5 Tumoren laufend über Jahre transplantiert: Liposarkom, Rhabdomyosarkom, Fibrosarkom, alveoläres Adenocarcinom der Speicheldrüse und ein solides Carcinom. Sie wachsen in den Recipienten jeweils subcutan, konstant schnell und mit gleichbleibender Histomorphe. Die Tumoren wurden parallel dazu in Zellkulturen gezüchtet. Die Subkulturen lassen sich in junge Ratten implantieren, wo sie wieder als solide Tumoren wachsen. An diesen Tumoren gleichartiger virogener Herkunft können spezielle Untersuchungen, wie DNS- und RNS-Isolierungen und Analysen durchgeführt werden.
