Summary
The oncogenic transformation of hamster kidney cells in vitro occurring over several weeks may be divided into 4 stages. The first stage is characterized by the appearance of sporadic multinucleated giant-cells. In the second stage one to five strongly refractile cell-islets appear in the cell sheet with increased numbers of nucleoli and increased cytoplasmic RNA among every 106 cells. In the third stage the strongly refractile islets of cell suddenly show numerous pathological changes: tri- and multipolar mitoses, highly increased DNA-metabolism, and by immunofluorescence, SV 40-specific T-antigen within cell nuclei. In the fourth stage epithelial cell formations develop surrounded by polymorphic spindle cells and multinucleated giant-cells. These epithelial cells also contain SV 40-specific T-antigen and possess a highly increased DNA-metabolism. The inoculation of transformed cells of the 3rd and 4th stage into Syrian golden hamsters induces malignant tumours that cause death.
Zusammenfassung
Der Vorgang der onkogenen Transformation von Hamsternierenzellen in vitro, der sich über mehrere Wochen erstreckt, läßt sich in 4 Stadien einteilen. Das 1. Stadium war durch das Auftreten vereinzelter, mehrkerniger Riesenzellen gekennzeichnet. Im 2. Stadium traten im Zellrasen mit einer Häufigkeit von 1–5 mal unter 106 Zellen stark lichtbrechende Elemente mit erhöhten Nucleolenzahlen und vermehrter RNS-Menge im Cytoplasma auf. Im 3. Stadium zeigten diese stark lichtbrechenden Zellinseln plötzlich eine Vielzahl von z. T. pathologischen, tri- und multipolaren Mitosen, einen sehr stark erhöhten DNA-Stoffwechsel und fluorescenzserologisch das SV 40-spezifische T-Antigen in den Zellkernen. Im 4. Stadium entwickelten sich epitheliale Zellverbände, die von polymorphen Spindelzellen und mehrkernigen Riesenzellen umgeben waren. Auch diese Zellen enthielten das SV 40-spezifische T-Antigen und hatten einen stark erhöhten DNS-Stoffwechsel. Die Verimpfung transformierter Zellen des 3. und 4. Stadiums auf Goldhamster führte zu bösartigen, tödlich verlaufenden Tumoren.
