Zusammenfassung

Bei einer Anzahl von Bakterienarten (Bacillus subtilis, Proteus vulgaris, Aerobacter aerogenes, Pseudomonas aeruginosa, Nocardia corallina, Corynebacterium michiganense, Rhizobium radicicola, Azotobacter agile, Mycobacterium phlei und Mycobacterium-Stämme: IVS-B, IBS-G, IBS-M) wurde geprüft, inwieweit die Entziehung von Kohlendioxyd die endogene Atmung, die Substratatmung und die oxydative Assimilation beeinflußt. Der CO2-Einbau wurde zur O2-Aufnahme in Beziehung gesetzt und als Quotient-ΔCO2/-ΔO2 angegeben. Eine Beziehung zwischen der Empfindlichkeit gegenüber dem Entzug von CO2 und der Größe des CO2/O2-Quotienten ließ sich nicht in allen Fällen feststellen.

Die höchsten Fixierungsquotienten (-ΔCO2/-ΔO2) wurden während der oxydativen Assimilation verzweigter Fettsäuren (Isovaleriansäure und Isobuttersäure) durch die neu isolierten Mycobacterium-Stämme IBS-G und IBS-M ermittelt.

Die Geschwindigkeit der CO2-Fixierung während der oxydativen Assimilation von Isobuttersäure durch Mycobacterium IBS-M nahm mit steigendem CO2-Partialdruck zu und erreichte bei 5% CO2 in der Luft einen Sättigungswert.

Die CO2-Fixierung durch Mycobacterium IBS-M mit Isobuttersäure als Substrat wurde eingehend untersucht. CO2 wurde zuerst vorwiegend in die nichtflüchtige, nichtätherlösliche Fraktion eingebaut; nach kurzer Einbauzeit (1–3 min) befand sich der Hauptteil des fixierten Kohlendioxyds in Asparaginsäure und Glutaminsäure.

Im zellfreien System mit Enzymextrakten aus Mycobacterium IBS-M wurden chromatographisch als erste Produkte der CO2-Fixierung mit Propionsäure als Substrat Bernsteinsäure und Methylmalonsäure, mit Isobuttersäure als Substrat, Bernsteinsäure und Dimethylmalonsäure nachgewiesen.

Es wird angenommen, daß der größte Teil des durch Mycobacterium IBS-M mit Isobuttersäure als Substrat fixierten Kohlendioxyds über die Carboxylierung von Propionyl-CoA eingebaut wird, und daß die direkte Carboxylierung von Isobutyryl-CoA in quantitativer Hinsicht lediglich einen Nebenweg darstellt.
