Summary
Conditions are described for converting l-ascorbic and dehydroascorbic acids to the trimethylsilyl derivatives for their gas-chromatographic separation. The silylation is carried out in non-aqueous acetone solution with a mixture of hexamethyldisilazane and trimethylchlorosilane. The silylation of dehydroascorbic acid according to Sweeley in pyridine solution results in the instantaneous formation of the enolate of 2.3-diketogulonic acid, the trimethylsilyl derivative of which is eluted together with that of l-ascorbic acid, so that no separation is possible in this way. The reaction rate of the silylation in acetone solution is remarkably smaller than in pyridine solution.
Zusammenfassung
Es werden Reaktionsbedingungen angegeben, unter denen man l-Ascor-binsäure und Dehydroascorbinsäure in die Trimethylsilyl-Derivate überführen und gas-chromatographisch trennen kann. Die Silylierung wird in wasserfreier, acetonischer Lösung mit einer Mischung von Hexamethyl-disilazan und Trimethylchlorsilan durchgeführt. Silyliert man die Dehydroascorbinsäure nach Sweeley in pyridinischer Lösung, dann entsteht aus ihr momentan das Enolat der 2,3-Diketogulonsäure, dessen TMS-Derivat gleichzeitig mit dem der l-Ascorbinsäure eluiert, also von diesem nicht getrennt wird. Die Reaktionsgeschwindigkeit der Silylierung in acetonischer Lösung ist deutlich kleiner als in pyridinischer Lösung.
