Zur Aussagekraft humanpharmakologischer Prüfmethoden für den Wirkungsvergleich von Corticoidexterna

Value of human pharmacology test methods in the comparative assessment of topical corticosteroids

Summary

In order to elucidate some fundamental questions of methodology in comparative pharmacological investigations in man, we studied the anti-inflammatory effect of 4 typical corticoids (1. Hydrocortisone, 2. Betamethasone, 3. Fluocinolone and 4. Flumethasone) in 4 experimental models of inflammation (1. Rubefacient Test, 2. Histamine-Dermatitis, 3. UV-Dermatitis and 4. Epidermal Allergy Contact Test) of the human skin. Since the effects of topical corticoids on different mechanisms of inflammation in the human skin vary considerably, the simultaneous employment of several experimental models proved to be a useful means of characterizing the pharmacological properties of topical corticoids.

Zusammenfassung

Zur Abklärung grundsätzlicher methodologischer Fragen bei humanpharmakologischen Vergleichsprüfungen haben wir den reaktionspräventiven Effekt von vier charakteristischen externcorticoiden (1. Hydrocortison, 2. Betamethasen, 3. Fluocinolon und 4. Flumethason) an vier artifiziellen Entzündungsmodellen (1. Rubefacienstest, 2. Histamin-Dermatitis, 3. UV-Dermatitis und 4. Epidermaler Allergen-Kontakttest) der menschlichen Haut studiert. Die gleichzeitige Benutzung mehrerer Entzündungsmodelle erwies sich dabei für die pharmakologische Charakterisierung von Corticoidexterna als sehr nützlich, da sich verschiedene Corticoide den differenten Pathomechanismen entzündlicher Hautprozesse gegenüber recht unterschiedlich verhalten.
