Summary
The permeability of the basal membrane and the intercellular space of the epidermis for low molecular weight proteins was studied in vivo. Peroxidase injected intracutaneously was used as electron microscopic tracer and the following observations were made:
-
1.
The basal membrane and the intracellular spaces of the epidermis including the desmosomes are readily and rapidly permeated by the tracer protein. Peroxidase can be demonstrated electron microscopically throughout the entire intercellular compartment of the epidermis.
-
2.
The intercellular spread of the tracer protein is stopped in the upper portions of the granular layer where lamellar structures derived from Odland-bodies block the intercellular spaces.
-
3.
Perinuclear cysternae were repeatedly observed which contained peroxidase. They are continuous with the intercellular space but their exact nature remains to be clarified.
It has been shown previously that a cement substance containing acid mucopolysaccharides occupies the intercellular space. The permeability of this compartment for low molecular weight proteins indicates that the intercellular cement possesses physico-chemical porperties which permit unrestricted access of such molecules to epidermal cells.
