Summary
Immunoelectrophoretic analysis of genuine seminal plasma with antisera specifically directed against human seminal plasma proteins reveals 12–13 different proteins. They are partly derived from the blood serum, partly they are “synthesized” by the accessory genital glands. The latter can be distinguished by using antisera against seminal plasma which have been completely absorbed with human serum proteins. Antisera which are specifically directed against human serum proteins may reveal further serum proteins which are present in the seminal plasma in very small amounts only.
These traces of proteins are enriched after chromatographic fractionation of seminal plasma on DEAE anion exchange cellulose and/or Sephadex G200 respectively followed by lyophilisation so that they can be demonstrated easier by immunological methods.
In this way we could demonstrate 9 “real” seminal plasma proteins and at least 10 serum proteins in human seminal plasma which are IgG, IgA, β1A/C-globulin, transferrin, α1-antitrypsin, α1-glycoprotein, orosomucoid and albumin as well as 1–2 proteins with the electrophoretic mobility of β1- and α2-globulins and another protein migrating faster than albumin.
With an antiserum, specifically directed against fragments of γ-globulins, we could demonstrate the presence of two γ-globulin fragments with molecular weights of approximately 30000 and 50000 respectively.
Furthermore, the isolation of two immunologically pure seminal plasma proteins has been performed for the first time by using gel filtration on Sephadex G 200 and preparative agar-electrophoresis. One of these proteins seems to derive from the prostate gland. On account of its sedimentation rate it has been called 1.1 S-seminal plasma protein (mol.-weight 8000–10000), the other one is probably synthesized by the epithelium of the seminal vesicles and has been designed as 3.72 S-seminal plasma protein (mol.-weight approx. 50000).
Zusammenfassung
Im nativen Spermaplasma können mit der immunoelektrophoretischen Analyse bei Verwendung von Antiseren, welche spezifisch gegen Spermaplasmaproteine gerichtet sind, 12–13 verschiedene Eiweißkörper nachgewiesen werden. Es handelt sich teils um Serumproteine, teils um Eiweißkörper, die in den akzessorischen Geschlechtsdrüsen gebildet werden. Letztere lassen sich mit Anti-Spermaplasma-Seren, welche mit Bluteiweißkörpern absorbiert wurden, gut unterscheiden. Mit Antiseren, welche spezifisch gegen menschliche Serumproteine gerichtet sind, können Bluteiweißkörper nachgewiesen werden, die im Spermaplasma nur in sehr geringer Menge vorkommen. Die chromatographische Fraktionierung über DEAE-Cellulose, bzw. Sephadex G 200 mit anschließender Lyophilisation führt zur Anreicherung solcher Eiweißspuren, so daß sie dem immunologischen Nachweis leichter zugänglich sind. Auf diese Weise konnten insgesamt 9 „echte” Spermaplasmaproteine und mindestens 10 Serumeiweißkörper nachgewiesen werden, und zwar IgG, IgA, β1A/C-Globulin, Transferrin, α1-Antitrypsin, α1-Glykoprotein, Seromucoid und Albumin, sowei 1–2 Proteine mit der elektrophoretischen Beweglichkeit von β1-, bzw. α1-Globulinene und ein Eiweißkörper, der schneller wandert als das Albumin.
Mit einem Antiserum, welches gegen γ-Globulin-Bruchstücke gerichtet ist, konnten 2γ-Globulinfragmente mit Molekulargewichten von ca. 30000, bzw. ca. 50000 nachgewiesen werden. Mit der Gel-Filtration über Sephadex G 200 konnten erstmals 2 Spermaplasmaproteine isoliert und in immunologisch reiner Form dargestellt werden; das eine stammt vermutlich aus der Prostata und wurde aufgrund seines Sedimentationskoeffizienten 1,1 S-Spermaplasmaprotein genannt (Mol.-Gew. 8000–10000), das andere wird in den Samenbläschen gebildet und wurde von uns als 3,72 S-Spermaplasmaprotein (Mol.-Gew. ca. 50000) bezeichnet.
