Summary
A new and sensitive method for the determination of substances containing alcoholic groups is based on the reaction with naphthylisocyanate to fluorescing urethanes. The wide applicability of this technique is demonstrated by means of the derivatisation of a great variety of alcohols with strongly differing structures. By applying triethylenediamine as a catalyst, primary and secondary alcohols may be converted into the corresponding carbamates within 30 min and tertiary alcohols within 2 h. Quantitative determination is performed by fluorodensitometric measurement of the highly fluorescent derivatives. By dipping the chromatogram into a methanolic solution of polyethyleneglycol an enhancement of sensitivity may be achieved up to a factor of 24. The detection limits determined in this way are on average in the lower pmole range.
Zusammenfassung
Mit der Umsetzung von Substanzen mit alkoholischer Hydroxylgruppe mit Naphthylisocyanat zu fluorescierenden Urethanen konnte ein neues, empfindliches Bestimmungsverfahren für diese Substanzklasse entwickelt werden. Die breite Anwendbarkeit der Methode wird anhand der Derivatisierung einer großen Palette strukturell stark unterschiedlicher Alkohole demonstriert. Durch Einsatz von Triethylendiamin als Katalysator können primäre und sekundäre Alkohole innerhalb von 30 min, und tertiäre Alkohole innerhalb von 2h in die entsprechenden N-Naphthylcarbaminsäureester übergeführt werden. Die quantitative Auswertung erfolgt durch fluorodensitometrische Vermessung der hochfluorescierenden Derivate. Durch Tauchen des Dünnschicht-Chromatogramms in eine methanolische Polyethylenglykollösung kann eine Empfindlichkeitssteigerung bis zum Faktor 24 erzielt werden. Die so ermittelten Nachweisgrenzen liegen im Durchschnitt im unteren pmol-Bereich.
