Summary
48 rats were treated with the antihypertensive drug, guanethidine; its effects on the parotid glands (α-amylase content, protein concentration, and isoamylase distribution patterns) were investigated. The treatment significantly increased the amylase activity in the gland extracts as well as the concentration of isoamylases and their specific enzyme activities. Moreover, we found a redistribution of activity between the 4 isoamylases of the parotid glands. The observed changes are the manifestations of a proteodyschylia concomitant with a peripheral neurogenic sialoadenosis. The described alterations are fully reversible within two weeks after discontinuation of the guanethidine application.
Zusammenfassung
Die Auswirkungen einer Guanethidinbehandlung auf die Ohrspeicheldrüse werden anhand des Amylasegehaltes und der Isoamylasenmuster in der Gl. parotis von 48 Ratten untersucht. Unter der Behandlung mit dem Antihypertonikum steigen die Amylaseaktivität im Drüsenextrakt, die Konzentration der Isoamylasen und deren spezifische Aktivität signifikant an. Zwischen den 4 Isoamylasen der Gl. parotis findet außerdem eine Aktivitätsverschiebung statt. Diese Veränderungen sind die Manifestation einer Proteodyschylie im Rahmen einer peripheren neurogenen Sialoadenose. Die Veränderungen sind innerhalb von 14 Tagen nach Absetzen des Medikamentes voll reversibel.
