Location and nucleotide sequence of the gene for the 15.2 kDa polypeptide of the cytochrome b-f complex from pea chloroplasts

Summary

The gene for the 15.2 kDa polypeptide of the pea cytochrome b-f complex has been located in pea chloroplast DNA by coupled transcription-translation of fragments of chloroplast DNA in a cell-free system from Escherichia coli. The gene is located 36 kbp away from the gene for cytochrome f and is transcribed from the opposite strand. Nucleotide sequence analysis of an 850 bp MspI fragment containing the gene reveals an open reading frame coding for a hydrophobic protein of 139 amino acid residues (M, 15266). A sequence of 50 amino acid residues in the middle of the 15.2 kDa polypeptide shows homology with a sequence near the C-terminus of mitochondrial cytochrome b.
