Haemodynamic effects of a new beta-adrenergic blocker (Trasicor®) in man when supine and sitting

Hämodynamische Wirkungen eines neuen ß-adrenergischen Blockes Trasicor® beim Menschen im Liegen und im Sitzen

Summary

The haemodynamic effects of CIBA 39 089-Ba (Trasicor) were studied in 17 subjects with normal or increased arterial pressure and marked differences in cardiac index. Studies were performed in both the supine and the sitting position. — In the supine position there were no changes in mean arterial pressure, a 3 % decrease in cardiac output, 5 % decrease in heart rate, a 2 % increase in stroke volume and little change in total peripheral vascular resistance after Trasicor administration (5 mg i.v.). In the sitting position the changes after Trasicor administration were about — 2% for mean arterial pressure, —10% for cardiac output, — 8% for heart rate, — 2% for stroke volume and — 8% for total peripheral vascular resistance. — The difference between Trasicor effects in supine and sitting positions is most probably due to the different amount of sympathetic nervous activity in these positions. The same may be true for the differences in effects between different subjects. — The haemodynamic effects of Trasicor are different from those of propranolol and resemble those of pronethalol.

Zusammenfassung

Die hämodynamischen Wirkungen von CIBA 39 089-Ba (Trasicor® wurden an 17 Versuchs-personen mit normalem oder erhöhtem arteriellem Druck und ausgeprägten Unterschieden des Herzminutenvolumenindexes studiert. Die Versuche wurden sowohl im Liegen als auch im Sitzen durchgeführt. — Im Liegen zeigten die Probanden nach intravenöser Gabe von 5 mg Trasicor keine Veränderungen des Blutdruekes, 3% Abnahme des Herzminutenvolumens, 5 % Abnahme der Pulsfrequenz, 2% Zunahme des Schlagvolumens und geringe Veranderungen des totalen peripheren Widerstandes. Im Sitzen betrugen die Veränderungen nach Gabe von Trasicor ungefähr — 2 % des arteriellen Druckes, —10 % des Herzminutenvolumens, — 8 % der Pulsfrequenz, — 2 % des Schlagvolumens und —8% des totalen peripheren Widerstandes. — Der Unterschied der Wirkung von Trasicor bei liegenden und sitzenden Probanden ist höchst wahrscheinlich durch die unterschiedliche Aktivität des Sympathicus bedingt. Das gleiche gilt vermutlich in bezug auf die Wirkungsunterschiede bei den verschiedenen Versuchspersonen. — Die hämodynamischen Wirkungen von Trasicor sind anders als die von Propranolol und entsprechen denen von Pronethalol.
