Abstract
A study has been made of the behaviour of glass-microelectrodes for intracellular measurements. The following facts have been obtained on a large number of experiments:
-
1.
The more negative the tip-potential of the electrode the greater the difference of the membrane potential from the actual value. For each 10 mV of the tippotential the membrane potential decreases by about 2.8 mV.
-
2.
The tip-potential is caused by the electrical charge of the inner surface of the elctrode-tip in connection with the concentration gradient between the solutions within and outside the electrode.
-
3.
The passage of the electrode from a high Na+- to a high K+-medium outside and inside the muscle cell respectively is only partly responsible for the error in the membrane potential values.
-
4.
It has been shown that the tip-potential of the electrode depends on the pH of the testsolution, i.e. the pH affects the charge on the inner surface of the glass-wall at the tip of the electrode. It is discussed that when the tip of the electrode is introduced into the cell, the electrical charge on the glass wall is affected by the electrical field of the membrane potential.
-
5.
For reducing the interfering tip-potential it is recommended to fill the electrode with 0.5-molar KCl-solution.
