Summary
The penetration through the epidermis in vitro of various topically used corticosteroids was compared. The amount penetrating through the epidermis from an ethanolic solution showed good correlation with the polarity of the corticosteroids under study. Hydrocortisone, the most polar corticosteroid, penetrates the epidermis the most rapidly; clobetasone butyrate, the least polar, the slowest. Other corticosteroids, i.e., hydrocortisone-17-butyrate, triamcinolone acetonide, and clobetasol-17-propionate, form an intermediate group whose penetration rates and polarities decrease in the indicated sequence. The corticosteroid generally accepted as having greater clinical efficacy in creams or ointments did not permeate better from an ethanolic solution in vitro.
Zusammenfassung
Die Permeation von fünf regelmäßig benutzten Corticosteroiden aus einer alkoholischen Lösung durch die Epidermis wurde in vitro gemessen. Diese Resorption zeigte eine gute Korrelation mit der Polarität. Hydrocortisone, das am besten permeiert, zeigt die höchste Polarität, Clobetasone-butyrat, dessen Polarität am geringsten ist, permeiert am wenigsten. Die weiteren untersuchten Steroide, wie Hydrocortisone-17-butyrat, Triamcinolone-Acetonid und Clobetasol-17-propionat, gehören zu einer mittleren Gruppe, in der Permeation und Polarität abnehmen, und zwar in der Reihenfolge in der sie genannt werden. Die Permeationsrate von solchen Corticosteroiden, die klinisch am besten wirksam sind, zeigt keine Parallelität im Hinblick auf bevorzugte Resorption.
