Das Akrosom bildet sich aus der Akrosomvakuole, die nahe der Kernoberfläche im Zusammenhang mit dem Golgikomplex entsteht. Der Ort ihrer Entstehung entspricht nicht der späteren Lage des Akrosoms. Neben dem Golgikomplex erscheinen während der Akrosombildung stark osmiophile Körnchen. Nach der Ausbildung des Akrosoms legt sich das Cytoplasma der Sertoli-Zellen der vom Akrosom bedeckten Kopfspitze der Spermatide eng an, und es treten Anzeichen eines Stoffwechsels zwischen Spermatide und Sertoli-Zelle auf.

Das Karyoplasma der jungen Spermatide hat die gleiche Struktur wie das anderer Interphasenkerne. Die schon während der Chromatinkonzentration sichtbaren, 180–200 Å dicken Körnchen werden in der 1. Phase gleichmäßiger verteilt und dichter. In der 2. Phase erscheinen grobe Körnchen von 600–700 Å an der Kernmembran; im Bereich der Kernkappe, und im kaudalen Kernbereich treten größere Vakuolen auf. In der 3. Phase werden die Körnchen im ganzen Kern vergrößert. Die grobe Granulation ist erst locker und wird zunehmend dichter. Schließlich ist das gesamte körnige Material bis auf Reste der weniger dichten Kernsubstanz zusammengebacken. In dieser Phase treten besonders große Vakuolen im kaudalen Karyoplasma auf.

Während der Chromatinkondensation treten Stoffe aus dem Kern aus. Schon in der 1. Phase wird elektronendichtes Material aus Membranporen abgegeben, die in regelmäßigen Abständen nebeneinander liegen. Die aus dem Kern abgegebenen Substanzen werden in Lamellen vom Kern abgehoben, wodurch „Lamellenkörper“ entstehen. Wenn die Kernvakuolen in der 3. Phase erscheinen, werden keine Lamellenkörper mehr gebildet. Die Flüssigkeitsvakuolen im Karyoplasma werden gegen Ende der Spermiohistogenese an das Cytoplasma abgegeben.

Das proximale Centrosom besitzt einen seitlichen Fortsatz, der schon von Meves (1899) beschrieben wurde.
