Summary
The effects of testosterone propionate on the mouse kidney were examined by means of light- and electron-microscopy. The application of androgen in male and female mice results in hypertrophy of the renal proximal tubule. As an equivalent of increased metabolism distinctive nuclear alterations are observed in the proximale tubule cell. These alterations are characterized by nuclear enlargement due to increased karyoplasmic hydration, by transmutation of heterochromatin into euchromatin, furthermore by folding of the nuclear membrane as well as by structural loosening of the nucleoli and a granule flow towards the nuclear membrane. The cytoplasm of the stimulated renal tubule cell shows a moderate increase in the hydration of the hyaloplasm which may be primarily due to the intensified protein synthesis. Increase in the number of the free ribosomes and the polysomes respectively was not definitely provable. Electromicrographes of kidneys from animals treated with testosterone during ten and sixteen days let suggest the initiation of a regulating mechanism (“wearing-off” phenomenon) limiting anabolization.
Zusammenfassung
Der Einfluß einer Testosteronbehandlung auf die Mäuseniere wurde in Varianten Versuchsreihen mit licht- und elektronenmikroskopischen Methoden untersucht. Unter der Androgenapplikation tritt beim männlichen und weiblichen Tier eine Hypertrophie des renalen Hauptstückepithels auf. Als Ausdruck einer gesteigerten Stoffwechselleistung sind an der Hauptstückepithelzelle charakteristische Kernveränderungen festzustellen, die sich durch Kernvergrößerung infolge vermehrter karyoplasmatischer Hydratation, durch die Umwandlung von Heterochromatin in Euchromatin, ferner durch die Einfaltung der Kernwand sowie durch die Strukturauflockerung der Nucleoli mit einem auf die Kernmembran gerichteten Granulastrom kennzeichnen. Das Zytoplasma der stimulierten Tubuluszelle weist eine mäßige Zunahme der Hydratation des Hyaloplasmas auf, die in erster Linie in Zusammenhang mit der erhöhten Eiweißsynthese stehen dürfte. Eine Vermehrung der freien Ribosomen bzw. der Polysomen ist nicht eindeutig nachweisbar. Elektromikroskopische Aufnahmen der Nieren von Tieren, die 10–16 Tage mit Testosteron behandelt wurden, lassen auf das Einsetzen eines die Anabolisierung begrenzenden Regulationsmechanismus („wearing-off“ phenomenon) schließen.
