Reference mutations for the β subunit of RNA polymerase

Summary

It is shown by the mixed reconstitution technique that in E. coli AJ7 rif-rJ7 rifampicin resistance resides in the β-subunit. Thus, the otherwise isogenic pair of strains AJ1 rif-s and AJ7 rif-rJ7 are suitable as a reference for genetic work on transcription. In addition, comparison of cyanogen bromide fragments of both strains by 2-dimensional electrophoresis exhibits a difference in one fragment probably due to charge density and not size, and therefore presumably an amino-acid exchange.
