Aphrophora salicina Goeze besitzt im ♂ diploid 28 Autosomen und X und Y.

Vor den Reifeteilungen finden konstant 5 Gonienmitosen statt.

Während den Spermatogonienmitosen treten zwei verschiedene Arten von Metaphaseplatten auf. In der letzten Spermatogonienmetaphase sind die homologen Chromosomen somatisch gepaart.

In der Diakinese tritt ein Auflockerungsstadium ein, während dessen die Chromosomen nur ganz schwach färbbar sind. Daran anschließend findet eine Wiederverdichtung der Tetraden statt.

Das X-Chromosom ist in der Metaphase I senkrecht zur Spindelachse orientiert. In der Anaphase I ändert es seine Lage und wandert ungeteilt und parallel zur Spindelachse zu einem der beiden Pole.
