The postnatal development of the ovary in the “nude” mouse

Summary

The postnatal development of the ovary of the heterozygous nude (nu/+) mouse with the genetic background BALB/c is very similar—if not identical—to that of other mouse strains. As other BALB/c mice, the nu/+ females become sexually mature during the 5th week post partum (p.p.). At this age the ovaries contain corpora lutea at various stages of differentiation.

In the ovaries of newborn and 1 week-old homozygous (nu/nu) mice, the differentiation of oocytes into primary and secondary follicles is delayed. In the third postnatal week, the ovaries of homozygous females contain more atretic follicles than those of their heterozygous littermates. This increased degeneration of follicles may account for the greater mass of secondary interstitial tissue, which is observed in the ovaries of adult nu/nu females. In nine out of ten of the 5 to 7 week-old nu/nu mice studied, the ovaries contained no-/or only very few corpora lutea. Thus in homozygous nude females, the onset of sexual maturity is delayed. This ovarial immaturity may persist throughout life. In other animals development may become normal.

In addition to the impaired postnatal development of the ovary, unspecific inflammation of the uterine wall (endo- and/or myometritis) was detected in 47% of nu/nu animals older than five weeks. No direct correlation was, however, found between the delay of sexual maturation and the inflammatory changes in the uterus as many of the animals with an endo-or myometritis possessed mature ovaries. The low fertility of the female homozygous “nude” mouse seems, therefore, to be caused not only by an impaired differentiation of the ovary but also by inflammatory processes in the uterus.

Zusammenfassung

Die postnatale Entwicklung des Ovars der heterozygoten “nude” (nu/+) Maus des Stammes BALB/c ist mit derjenigen anderer Mäusestämme vergleichbar. In der 5. Woche post partum (p.p.) lassen sich erstmals unterschiedlich ausgereifte Corpora lutea im Ovar nachweisen; d.h. die nu/+ Weibchen werden zur gleichen Zeit geschlechtsreif wie andere Tiere ihres Stammes.

Bei der homozygoten “nude” (nu/nu) Maus ist bis zur zweiten Lebenswoche in den Ovarien eine verzögerte Umwandlung von Eiballen in Primärfollikel zu erkennen. In der 3. Woche p.p. weist das Ovar der nu/nu Maus deutlich mehr atretische Follikel auf, als dasjenige des nu/+ Tieres. Durch diese gesteigerte Atresie könnte die bei älteren nu/nu Weibchen beobachtete Zunahme des sekundär-interstitiellen Gewebes erklärt werden. 9 von 10 untersuchten nu/nu Weibchen ließen in der 5. bis 7. Woche p.p. keine — oder nur sehr wenige Corpora lutea in ihren Ovarien erkennen; d.h. diese Tiere waren noch nicht geschlechtsreif. Die in dieser Altersstufe unvollständige Differenzierung des Ovars kann sich entweder im Erwachsenenalter normalisieren oder aber bestehen bleiben.

Neben den Differenzierungsstörungen des Ovars wurde bei 9 von 19 nu/nu Weibchen, die älter als 5 Wochen waren, eine Endo- und/oder Myometritis beobachtet. Es konnte jedoch kein direkter Zusammenhang zwischen dem Vorliegen einer Infektion des Uterus und der verzögerten bzw. fehlenden Ausreifung des Ovars nachgewiesen werden, da ein Teil der nu/nu Weibchen mit einer Endo- bzw. Myometritis altersentsprechend entwickelte Ovarien besaß.

Die geringe Fertilität der weiblichen, homozygoten “nude” Maus ist sowohl auf die verzögerte Ausreifung des Ovars als auch auf die häufig auftretenden Infektionen des Uterus zurückzuführen.
