Fermenttopochemische Untersuchungen an der Nebennierenrinde des Meerschweinchens nach Metopironzufuhr

Summary

The histotopoohemical reaction of some enzyme systems of the adrenal cortex was studied in 76 male Guinea-pigs during the administration of metopirone. The results may be divided into 3 groups:

  1. 1.

    No changes are observed in esterases and acid phosphatases.

  2. 2.

    During the initial phase of the metopirone treatment alkaline phosphatases, adenosinetri-phosphatase, NAD-cytochrome-c-reductase and succinate dehydrogenase show increased activities in the zona fasciculata. On the 14th day, however, the activities are considerably lower as compared to the controls. Alkaline phosphatases reach subnormal values on the 28th day only. A general marked decrease in activity is noted on the 84th day.

  3. 3.

    The activity of glucose-6-phosphatedehydrogenase and 3β-ol-steroiddehydrogenase is reduced 24 hours after the first administration of metopirone. In the following period the activity of both enzymes increases gradually. Here too a subnormal enzymatic activity in the zona fasciculata is observed on the 84th day.

Zusammenfassung

An 76 männlichen Meerschweinchen wurde das histotopochemische Verhalten einiger Fermentsysteme der Nebennierenrinde während Metopironzufuhr untersucht. Die Befunde lassen sich in 3 Gruppen einteilen:

  1. 1.

    Die Esterasen und sauren Phosphatasen zeigen keine Veränderung.

  2. 2.

    Die alkalischen Phosphatasen, die Adenosin-tri-phosphatase, die NAD-Cytochrom-c-reductase und die Succinodehydrogenase zeigen zu Anfang eine erhöhte Aktivität in der Zona fasciculata, am 14. Tag jedoch eine deutliche Aktivitätsabnahme gegenüber den Kontrollen. Die alkalischen Phosphatasen fallen allerdings erst am 28. Tag auf subnormale Werte ab. Am 84. Tag wird allgemein ein starker Aktivitätsschwund festgestellt.

  3. 3.

    Die Glucose-6-phosphatdehydrogenase und die 3β-ol-Steroiddehydrogenase zeigen 24 Std nach der ersten Metopirongabe einen Abfall der Aktivität. Im weiteren Verlauf nimmt bei beiden Fermenten die Aktivität in der Zona fasciculata wieder zu. Am 84. Tag konnte auch hier eine subnormale Fermentaktivität in der Zona fasciculata nachgewiesen werden.
