Zusammenfassung
1. In Suspensionen isolierter chromaffiner Granula des Nebennierenmarks von Rind und Schwein erfolgt bei der Inkubation (37° C) in Luftoder Stickstoff-Atmosphäre schon spontan eine Freisetzung von Adrenalin und Noradrenalin. Diese spontane Freisetzung läßt sich durch Zugabe indirekt wirkender Sympathicomimetica, z.B. Tyramin, β-Phenyläthylamin, Benzedrin, Pervetin und Ephedrin, dosisabhängig verstärken.
Tyramin und β-Phenyläthylamin verursachen eine vermehrte Freisetzung von Noradrenalin auch aus isolierten Granula sympathischer Nerven (Milznerven).
Aus diesen Ergebnissen geht hervor, daß die sogenannte „indirekte“ Wirkung des Tyramins und anderer Amine tatsächlich in einer Freisetzung granulär gespeicherten Adrenalins und Noradrenains bestehen könnte.
2. Der physiologische „Freisetzer“ der Nebennierenmarkhormone — Acetylcholin — vermag ebensowenig wie Nicotin und Histamin aus isolierten Granula Adrenalin und Noradrenalin freizusetzen.
Die in vivo durch diese Pharmaka erfolgende Aminfreisetzung kann deshalb ihren Angriffspunkt nicht an der Membran der granulären Aminspeicher haben, sondern vermutlich an der Membran der granulahaltigen Zellen.
3. Cocain hemmt weder die spontane Aminfreisetzung aus den Granula, noch die durch Tyramin verstärkte Freisetzung.
Die in vivo durch Cocain ausgeübte Hemmung der Tyraminwirkung hat deshalb ebenfalls ihren Angriffspunkt wahrscheinlich nicht an der Granulamembran, sondern an der Zellemembran, in dem durch eine „membrandichtende“ Wirkung dem Tyramin der Weg zu den intracellulären Hormonspeichern verlegt wird.
Summary
1. The indirect sympathomimetic action of tyramine and of other amines is assumed to be mediated by a liberation of noradrenaline and adrenaline. In the suprarenal medulla as well as in sympathetic (splenic) nerves, these catechol amines are mainly stored in specific elements of the cell, the chromaffin granules.
2. When isolated medullary granules from cattle or pig suprarenals are suspended in isotonic sucrose-phosphate buffer ofph 6,8 and incubated at 37° C in air- or in N2-atmosphere a release of the stored catechol amines can be observed. This spontaneous release was found to be accelerated by addition of the indirectly acting sympathomimetic amines tyramine, β-phenylethylamine, amphetamine, metamphetamine, and ephedrine. The directly acting amine dopamine was ineffective.
3. The physiological „liberator“ of the medullary hormones — acetylcholine — as well as nicotine and histamine were found to be unable to release the hormones from isolated granules.
4. Cocaine inhibited neither the spontaneous amine release nor the enhanced released caused by tyramine from medullary granules.
5. The isolated granules from sympathetic nerves released spontaneously noradrenaline during the incubation period. Tyramine and β-phenylethylamine accelerated also this release, whereas dopamine did not.
6. These results support the hypothesis that the actions of tyramine and other indirectly acting sympathomimetic amines are mediated by a release of noradrenaline and adrenaline from chromaffin granules. — The failure of acetylcholine, nicotine, histamine, and cocaine to act on these granules suggests that the site of action of these compounds is probably the cell membrane.
