An improved lithium acetate method for yeast transformation

Summary

We have studied the effect on Saccharomyces cerevisiae transformation frequency of varying several parameters of the lithium acetate-mediated transformation protocol first reported by Ito et al. (1983 a). We found that by sequentially optimising these conditions we obtained an approximately 50-fold increase in yield from 190 to 9000 transformants per μg of the plasmid YEp 13 for strain DBY 746.
